http://news.yahoo.com/s/afp/20071026/hl_afp/healthdiseaseaids Fri Oct 26, 4:17 AM ET PARIS (AFP) - A prototype drug, tested on lab-dish samples of the AIDS virus, has shown great promise in attacking HIV from an unprecedented direction, French researchers reported on Friday. The drug inhibits the human immunodeficiency virus (HIV) at a key point -- when it uses the hijacked machinery of an infected immune cell to reassemble its genetic code. This process, called splicing, joins together sections of viral code to form pre-messenger RNA, which is a precursor for messenger RNA (mRNA). This molecule provides the blueprint for cranking out viral proteins and enzymes which are essential for replication. Once these are created, baby viruses can be reproduced en masse within the cell, eventually breaking out and heading into the bloodstream, where they go on to infect other cells. The scientist found that a test molecule, called IDC16, interferes with a human splicing protein called SF2/ASF that is used to do the pre-messenger RNA cut-and-paste. As a result, the reproduction process was braked at the start. In lab-dish tests, IDC16 blocked replication of different strains of HIV-1, including samples from patients who are resistant to the famous "cocktail" -- combination therapy -- of anti-HIV drugs. The paper appears in the US open-access journal Public Library of Science (PLoS) Pathogens. In a press release, France's National Centre for Scientific Research (CNRS), which is backing the project, said IDC16 was "extremely promising." The "cocktail," first introduced in 1995, has been a lifeline for millions of people by attacking the protein gp120, which is on the surface of the virus, and enzymes that are used in other phases of the replication process. The problem, though, is that HIV mutates, making these proteins a shifting target for drug engineers. The authors, led by Jamal Tazi of the Institute of Molecular Genetics in Montpellier, are confident that IDC16 throws up a way of sidestepping mutation. "Instead of attacking the components brought along by the virus, we use the machinery that it uses in the cell," Tazi told AFP. By aiming at splicing, other viruses could be targeted, Tazi added. "These findings may serve as the basis for a new strategy to develop a new class of anti-HIV drugs, the splicing inhibitors, and even of antiviral drugs in general, since any virus needing to splice its RNAs may be targeted," his team writes. However, exhaustive tests on lab animals, leading to cautious trials on humans, are needed before IDC16 can be certified as safe and effective. This process typically takes years."

When Viruses Become Hijackers
While there is still no cure for HIV, scientists have learned much about the virus and how to treat it since the first U.S. cases were identified in the early 1980s. Among those helping to expand the knowledge base is Melissa Nashat, an assistant professor in the BMCC department of science. Prior to joining the BMCC faculty in 2005, Nashat was a postdoctoral fellow at Albert Einstein College of Medicine Department of Pathology, where she analyzed thinly sliced sections of brain tissue from HIV patients under the mentorship of neuropathologyist Sunhee C. Lee. "We were looking for specific markers for HIV, including the p24 protein, which is present only in brains inflamed by HIV encephalitis," she says. Building "viral factories" Somewhat to their surprise, Nashat and her colleagues at Einstein found that uninflamed HIV brains -- those where the p24 protein is absent -- "actually bear more resemblance to normal, healthy brains than they do to ones with HIV but no inflammation." Equally unexpected, the researchers observed that in p24+ brains, infected cells do not typically die, but are instead "hijacked by the virus to become viral factories, causing the inflammation to worsen," Nashat says. Ideally, these findings will help shed light on the pathways through which the brain is affected by HIV, ultimately leading to pharmacological approaches to controlling the inflammation, which often results in HIV-associated dementia. One of the main challenges of HIV research is that "the virus is like the dust in your house -- it can hide out in the cells, defying detection, so that no matter how aggressively you treat it, you can never get it all," Nashat says. "The virus actually integrates its own DNA into the body's cells." Notwithstanding the tenacity of the virus, medicines that inhibit its proliferation have given new hope to HIV-positive individuals. "But the reality is that HIV is smart and learns to mutate quickly -- often into a form that is resistant to the drugs," she says. "As a result, a patient may get better for a while only to have the virus rebound and come back." Currently in clinical trials are drugs known as integrase inhibitors that are designed to prevent the virus from integrating its DNA, and tampering with the patient's genetic information. Universal interest Nashat, who teaches biology for non-science majors as well as more advanced courses for science majors and nursing students, continues to pursue her research -- and answers to the riddle of HIV-associated dementia -- at Einstein. At BMCC, she finds that students in all her classes are extremely interested in learning about HIV. "With science and nursing majors, I can go into some detail about the virus and the ways it causes brain inflammation and dementia," she says. "But non-science majors are also hungry for information.
